The Cell and Gene Bank will provide a central repository for the distribution of reference cell lines and HLA genes to support the scientific projects and core components of the 13th International Histocompatibility Workshop. The repository will include a comprehensive inventory of cell lines and HLA genes available to the IHWC and HLA community. Many of the reference cell lines in inventory will be derived from previous International Workshops. Reference cells from HLA heterozygous and homozygous donors, from hematopoietic cell transplant (HCT) patients and donors, from individuals representing selected population groups and from individuals expressing potential HLA null alleles will be acquired as available. Current Activity. The inventory of the Cell and Gene Bank is currently being established and expanded. At present the inventory comprises more than 75 B-LCL from the 10th IHWC, 90 B-LCL from selected CEPH families and 10 B-LCL representing null alleles. The remaining LCL from the 10th IHWC, as well as selected LCL from the 12th IHWC, will be expanded and placed in inventory over the next few months. Additional cell lines from local and international repositories are being requested as needed. Selected cell lines from over 500 HCT pairs will also be placed in inventory. Twenty HLA class I genes (e.g., A*0201; B*0702; C*0401), available in pCR2.1 or GEM-T cloning vectors and pNS or pcDNA3.1 expression vectors, are currently in the Gene Bank inventory. A current listing of cells and genes available from the bank is posted here. Reference Cells. The reference cell panels to monitor amplification and hybridization in SSOP, SBT and reverse line blot assays are now available. Order forms are on-line. Individual component projects using these reference cells for their respective studies will also announce their availability. Please consult specific component pages on this web site for further information. Cells Needed. We are interested in hearing from anyone with cells or HLA genes to contribute to the Cell and Gene Bank (see "Cells Needed") . If you have peripheral blood mononuclear cells or B-lymphoblastoid cell lines from individuals expressing HLA class I or class II genes of interest, or HLA genes (cDNA or genomic DNA) cloned in plasmid vectors available, please contact Eric Mickelson at: phone - 206-667-4922, fax - 206-667-6969, or email - emickels@fhcrc.org. Special Thanks. -- to Dr. Ee Chee Ren, Singapore and Dr. Yoshihide Ishikawa, Tokyo for their generous support in providing cells and cell lines to the Bank. Also thanks to Dr. Yoshiki Akatsuka for providing cloned class I genes and HLA transfected cell lines. |
