The number of participants in the SSOP component is low based on the number of oligonucleotide orders from LifeCodes and on the sets of QC samples requested. No typing data has been received. A letter requesting an update has received no response. The organizers will contact those participating to determine their status.
The organizing committee should submit their primary typing data using workshop reagents on the 52 cell reference panel and the QC cell panel to the data analysis component. We should aim for at least two labs performing the assay on each locus and each strategy (allele/registry) for each cell. The major emphasis is on A/B/C but class II will be collected if available.
The reagents which now differ from new alleles in proposed noncritical regions (5' and 3' edges of probes and 5' of primers) will be identified by Helmberg. These reagents will be flagged in the web/printed documents. The criteria for probes will be maximum of 2 nucleotides total at 5' and/or 3' edges. Mismatches in 5' 9 nucleotides of primers will be identified. Helmberg will evaluate this based on current set of sequences (all loci) and on the December 98 sequence list for class II. A comment will be added to the introduction to explain this issue by Hurley.
Helmberg will provide the typing strategies from his SCORE program so that the organizing committee can confirm that the program and the written material are consistent. This review will also confirm the alleles detected by the reagents using the December 1998 and October 2000 allele sets as printed out by Helmberg. Members of the organizing committee will review the documentation for accuracy one last time and add any comments on conditions.
Hurley will prepare an excel spread sheet listing comments about the quality of the SSOP reagents. Some organizers have already submitted information to her; others should send if available.
