Cytokine gene polymorphisms have increasingly become a topic of interest in laboratories that traditionally have engaged in histocompatibility testing and transplantation. This interest is due to the fact that certain of these polymorphisms have been shown to correlate with cytokine plasma levels in vitro or with the survival of kidney, heart or lung allografts. We therefore felt that it would be appropriate and desirable to establish a cytokine polymorphism component (CPC) within the framework of the 13th IHWG. Since there is little information available about the genetic diversity of these genes in different ethnic groups, it was suggested that anthropological studies should be included in the CPC project. It was also proposed that typing for cytokines would be an interesting topic for study within in the hematopoietic stem cell transplant component

Goals
The Cytokine Polymorphism project will study the genomic variability that characterises genes encoding certain cytokine molecules. The following 15 cytokine genes have been identified as candidates for study in this component: IL1 alpha; IL1 beta; IL1R; IL1RA, IL2; IL4; IL4R; IL6; IL10; IL13; IL15; TGF beta 1; TNF alpha; TOL-R; and gamma-IFN.

Research Plan
The Research Plan for this project will divide the study into two main phases. Phase I will be a preliminary "pre-workshop" project to assess the current state-of the-art technology, to obtain objective data to determine the performance of existing methods and reagents, and to determine the extent to which results can be replicated in different laboratories. In this phase, all participating laboratories will be sent a common set of approximately 50 coded DNA samples. These will be analyzed using available local reagents, using a set of pre-dispensed PCR-SSP reagents from the University of Heidelberg (provided at cost), or using commercial reagents. Participating laboratories may choose to test both their local reagents as well as the Heidelberg and/or commercial reagents.

Phase II studies. Following phase I, the organizing committee will choose a standard set of reagents to be used by all laboratories for phase II studies. The final protocol for this has not been completed, but we anticipate that each participating laboratory will carry out phase II studies on at least 100 samples collected from selected racial/ethnic groups, transplant patients, or patients being studied in the disease components.

Data Submission
To submit cytokine gene polymorphism data for samples tested with the SSP assay or other methods, please proceed as follows:

If you use the Cytokine-enhanced SCORE (will work only with the "Heidelberg PCR-SSP Cytokine Set") - Enter all reaction patterns in SCORE and Export all data you would like to share by using the "Export data" Option in Score (For transfer use the file: "transferLABCODEDATE.vda", which is located in your Score root-directory. Select the samples you want to transfer. This will generate a file with the name: cytokineLABCODEDATE.xls. This file contains all information, which is required for the evaluation of the cytokine data.

If you don't use SCORE use the Phase I or Phase II Data Capture Forms.

To submit data for the joint HCT-CGP project, please use the new combined data forms.

To submit demographic information on these samples, please use the Anthropology Data Submission Form. All data will be reported to the central database. Data analysis will be carried out by participating laboratories and members of the organizing committee according to their individual interest. The preliminary time schedule of the project is as follows:
