January 5, 2000

Dear Colleague,

I enclose herewith a follow up questionnaire on the HLA and Mycobacterial Disease component of the forthcoming 13IHWC. Please fill up and return to us latest by 30th January ,2000. We are making efforts to have an excellent study done so that meaningful data can be generated . Fortunately the CNG group in France has agreed to perform micosatellite typing on a battery of 11 microsatellites for the disease component of the workshop , We have made a request to them to include mycobacterial diseases for microsatellite typing.

The following needs to be carried out by you:

1. Please conduct HLA class II typings (low/intermediate/high ) for DRB1, B3,B4,B5 and DQB1 alleles, for all the samples that you submit . We would then conduct confirmatory typings and high resolution typings where ever needed. We are also trying to include sequence based typing, if possible.

2. Please ensure to save and send us 50 mg DNA for microsatellite studies including Nramp1. The DNA should be sent in 1.5 ml screw capped tubes with proper labeling. It may be noticed that good quality DNA is necessary to perform all of the above tests.

3. Also, please ensure to collect and send sera samples for sCD23 analysis. It will be appreciated if you could collect sera at 0,3 and 6 months time after treatment. If you are able to do so, please collect sera at one point time at least.

4. For the cytokine study by flowcytometry, you may isolate, fix ,freeze and ship the lymphocytes as per the protocol that has already been communicated to you.

5. It is important to include ethnically matched healthy controls in the study.

6. Please acknowledge our emails/faxes/letters both at (narin98@hotmail.com , nkmehra@medinst.ernet.in) so that we are assured that you have received the desired information in time.

With regards and best wishes for the millennium year 2000.

Prof. N.K.Mehra, Chair
